Her manager, Ron Richardson, said the star had been ill for some time, and finally passed away at a Chicago hospital on Monday night.
Beginning her career by singing in gospel choirs in her native Chicago, Loleatta recorded soul albums in the early 70s, before lending her voice to disco tracks later in the decade.
It was the 1980-hit Love Sensation that really launched Loleatta as an international star, and provided the singer with a windfall when the vocals were sampled by Black Box for their 1989 UK number one Ride on Time.
The song was also sampled by Donnie and Mark Wahlberg in the Marky Mark and the Funky Bunch single Good Vibrations.
Boy George led the tributes to Loleatta on Twitter, with the former Culture Club star saying: 'God rest your soul Loleatta Holloway, you were the greatest voice and I loved you! R.I.P.'
Other musicians have also paid their respect to the star, with DJ Kissy Sellout tweeting: 'R.I.P Loleatta Holloway. A rare talent who held such extraordinary importance to the history and influence of house music.'
Although no formal funeral plans have been made as yet, her manager revealed that a public memorial will be held in the singer's honour.
